loqs promoter and regulatory regions (2.5 kb upstream and 0.8 kb downstream of the locus) drive expression of a cDNA encoding the loqs-PA isoform with a 3xT:Zzzz\FLAG flag added to the amino-terminus of the protein. The construct is in a pattB vector allowing site-specific insertion into the genome.

loqs[PA.T:Zzzz\FLAG] restores the embryonic viability of loqs[KO] mutants. The rescued males are fertile, however, females lay far fewer eggs than wild-type controls. None of the eggs produced by loqs[KO] mutants rescued by loqs[PA.T:Zzzz\FLAG] hatch, and few of the eggs display normal dorsal appendages. loqs[KO] mutant females rescued by loqs[PA.T:Zzzz\FLAG] together with loqs[PD.T:Zzzz\FLAG] lay far fewer eggs than wild-type controls. None of the eggs produced by the rescued females hatch. Few of the eggs laid by the rescued females display normal appendages. loqs[KO] mutant females rescued by either loqs[PA.T:Zzzz\FLAG] or the combination of loqs[PA.T:Zzzz\FLAG] and loqs[PD.T:Zzzz\FLAG] display fewer germline stem cells (GSCs) per germaria than wild-type controls.
