leg, with Scer\GAL4Dll-md23
Expression of discodsRNA.Scer\UAS using Scer\GAL4αTub84B.PL causes lethality at both larval and pupal (but not embryonic) stages.
Expression of discodsRNA.Scer\UAS using Scer\GAL4Dll-md23 causes some lethality but allows many flies to survive. At 25[o]C, ~64% of animals survive; all escapers show antenna phenotypes characterized by shortening of the central core and deletion of most of the laterals; leg development is also defective but only 10% show deletion of the distal tarsal segments and/or the claw. When raised at 29[o]C for 4-5 days and then transferred to 25[o]C, the survival rate decreases to ~29%; now, over 70% escapers display antenna to leg transformations and the remainder display partial truncation of antennal segment 3 and arista deletion; the leg phenotype is also more severe with ~16% of escapers displaying deletion of tarsal segments 3-5 and the claw.
Expression of discodsRNA.Scer\UAS using Scer\GAL4Dll-md23 does not induce cell death in antennal or leg discs.
