UAS regulatory sequences drive expression of an inverted repeat of a 443bp step cDNA fragment (basepairs 589-1031 of step-RA cDNA, corresponding to the C-terminal pleckstrin homology domain) that forms a dsRNA fragment.

stepdsRNA.Scer\UAS-mediated knockdown of step in the posterior wing compartment, under the control of Scer\GAL4en-e16E, results in a reduction of the wing size due to a reduction in cell number but not size. Furthermore, loss of wing vein tissue is observed and both cross veins are not formed properly. Reducing step levels further by RNAi mediated knockdown in a Df(2L)TW65 background enhances the wing vein phenotype with absent crossveins and gaps in the lateral vein L4.

The reduction of wing vein tissue seen upon expression of cnkGD11960 in the posterior compartment of the imaginal wing disc under the control of Scer\GAL4en-e16E can be further increased by co-expression of stepdsRNA.Scer\UAS.

Reducing Egfr levels by EgfrKK100051-mediated knockdown in the posterior wing compartment (under the control of Scer\GAL4en-e16E), in a stepdsRNA.Scer\UAS background generates a more severe Egfr phenotype. The lateral vein L4 is entirely lost, both cross veins are not differntiated and the distal end of the lateral vein L5 is missing.

Knockdown of Ras85D in a stepdsRNA.Scer\UAS background, through expression of Ras85DKK108029 in the posterior wing compartment (both transgenes under the control of Scer\GAL4en-e16E) generates a more severe Ras85D phenotype than in a wild-type background. The lateral vein L4 is entirely lost, both cross veins are not differentiated and the distal end of the lateral vein L5 is missing.
