UAS regulatory sequences drive expression of a Zzzz\CAG-repeat encoding 55 glutamines. The repeats are flanked by six amino acids on the N-terminal side and four on the C-terminal, and the Tag:MYC and Tag:FLAG epitope tags are located downstream.
Expression of one copy of Zzzz\CAG55.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4GMR.PFa results in mild eye pigmentation loss, localised mainly to the posterior of the eye. Very few photoreceptors are evident in the ommatidium. The phenotype is more severe when two copies of the transgene are expressed.
Brains from adult flies expressing Zzzz\CAG55.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4elav.PLu exhibit a large number of aggregates that are localised to the neuronal cell body. They are found in the central brain, the optic lobes nd the antennae. A single aggregate is seen per nucleus.
