UAS regulatory sequences drive expression of Hsap\HUWE1.
Flies expressing two different insertions of Hsap\HUWE1Scer\UAS.cVa (PBac{UAS-HUWE1.V}VK00031 and PBac{UAS-HUWE1.V}VK00037) under the control of Scer\GAL4Mef2.247 do not show any learning or short term memory defects when subjected to a courtship conditioning paradigm. When expressed in the γ and some α/β neurons of the mushroom body under the control of either Scer\GAL4Tab2-201Y of Scer\GAL4Mef2.247 no aberrations are seen in the axon lobes. The calyx also appears normal.
No morphological defects are seen in the NMJ of muscle 4 in wandering third instar larvae when Hsap\HUWE1Scer\UAS.cVa is expressed under the control of Scer\GAL4elav.PU (co-expressed with Dcr-2Scer\UAS.cDa). The NMJ area, perimeter and length are unaffected, and there are no differences in the number of branches and branching points. The number of active zones is also unaffected.
Adult flies expressing Hsap\HUWE1Scer\UAS.cVa under the control of Scer\GAL4nSyb.PS do not present any abnormalities in their electroretinogram (ERG) profile compared to controls.
When Hsap\HUWE1Scer\UAS.cVa is expressed in the dorsal cluster neurons under the control of Scer\GAL4ato.3.6 the number of axons crossing the optic chiasm is similar to controls. However, the branching pattern in the adult medulla is disturbed, giving rise to an increased number of branches at the 3rd branching point of the grid-like structure.
Hsap\HUWE1UAS.cVa, Scer\GAL4ato.3.6 has abnormal neuroanatomy | adult stage phenotype, suppressible | partially by armS10.UAS.Tag:MYC, Scer\GAL4ato.3.6
Hsap\HUWE1UAS.cVa, Scer\GAL4ato.3.6 has lobula columnar neuron LC14 | adult stage phenotype, suppressible | partially by armS10.UAS.Tag:MYC, Scer\GAL4ato.3.6
Hsap\HUWE1UAS.cVa, Scer\GAL4ato.3.6 has medulla | adult stage phenotype, suppressible | partially by armS10.UAS.Tag:MYC, Scer\GAL4ato.3.6
Expression of armS10.Scer\UAS.T:Hsap\MYC partially rescues the dorsal cluster neuron branching phenotype seen when Hsap\HUWE1Scer\UAS.cVa is expressed under the control of Scer\GAL4ato.3.6.
