Description

Mutation of the three putative microtubule tip localisation sequence (MtLS) motifs in the Ctail region. In each case the putative MtLS motif is replaced by four Ala residues. The location of each MtLS motif is as follows: amino acid residues 4966-4969, 5077-5080 and 5141-5144 (coordinates numbered according to FBpp0086744).

The expression of shotRE-3MtLSmut.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4en.PU rescues the abnormal microtubules observed in the dorsal-most epithelial cells of shotSF20 homozygous embryos during dorsal closure.

Expression of shotRE-3MtLSmut.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4eve.RN2O does not rescue the motor neuron stall phenotype seen in shotunspecified embryos in vivo and expression under the control of Scer\GAL4sca-537.4 fails to rescue the axon extension and microtubule organisation defects of shotSF20/Df(2R)MK1 embryonic neurons in primary culture.

Expression of shotRE-3MtLSmut.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4sr-md710 restores tendon cell integrity in shotunspecified embryos.
