The NMJs of larvae expressing mnbScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4nSyb.PS show an increased number of synaptic boutons compared to controls, and these boutons are smaller than in controls.
Expression of mnbScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4nSyb.PS has no effect on synaptic transmission or responses to high-frequency stimulation compared with controls.
Larvae expressing mnbScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4nSyb.PS exhibit enhanced synaptic vesicle endocytosis: the level of membrane uptake (dye internalisation) in mutant synaptic boutons in response to nerve stimulation is significantly increased compared to controls. When dye unloading occurs in response to a second stimulation, the mutant larvae also unload an increased amount of dye to controls.
Pre-synaptic expression of mnbScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4nSyb.PS rescues the bouton and miniature excitatory postsynaptic potential (mEPSP) defects seen in mnb1 mutants. The synaptic vesicle endocytosis defects are also rescued.
