The ple promoter region is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.

Inhibition (using shi1.UAS at 30-33[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.ple and Hsap\RELAAD.R52H03 in adults, only during aversive (CS+) odor re-exposure following training, does not prevent the aversive olfactory memory extinction that results from re-exposure, compared to controls.

Adult females prefer optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 627 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.ple and Hsap\RELAAD.R52H03 to darkness with no activation; this preference is significantly greater than that of controls for red light compared to darkness.

Inhibition (using shi1.UAS at 33[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.ple and Hsap\RELAAD.R52H03 in adults significantly reduces aversive, but not appetitive, 1-min visual memory, compared to controls; inhibition does not significantly affect shock preference, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 31[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.ple and Hsap\RELAAD.R52H03, in combination with a paired light stimulus, results in a significant increase in short-term associative aversive visual memory, compared to controls.
