The P{UAS-mCherry.mir-276a.sponge.V2} and P{UAS-mCherry.mir-276b.sponge.V2} constructs target mir-276a-3P and mir-276b-3P respectively. However, because the only difference between mir-276a-3P and mir-276b-3P is at position 10, and position 10 is in the sponge bulge (not complementary to the miRNA), both sponges target both miRNAs.
Adult-onset expression of mir-276aa.sponge.V2.Scer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4Pdf.Switch1a (used with RU486-supplemented diet to activate the expression in adult flies only) results in arrhythmic circadian activity in 44% of adult flies. Circadian free-running period in the mutant flies that remain (at least partially) rhythmic is higher than in controls, but most likely just due to inaccuracy of the measurements caused by the decreased rhythmicity.
The lack of a Scer\GAL4tub.PU-mediated viability phenotype is a false negative (compared to independent viability data from miRNA deletion mutants).
