Flies with expression of U2AdsRNA.shRNA.Scer\UAS.P\T driven by Scer\GAL4αTub84B.PL only during adulthood (with TubGAL80ts put at 29[o]C after eclosion for 7 days) leads to complete male sterility. Expression of U2AdsRNA.shRNA.Scer\UAS.P\T driven by Scer\GAL4nos.UTR.T:Hsim\VP16 leads to reduced fertility in males (but not females); expression driven by Scer\GAL4C587 (in adulthood with TubGAL80ts) or Scer\GAL4upd1-E132 has no effect on fertility, whereas expression driven by Scer\GAL4bam.T:Hsim\VP16 leads to complete male sterility and defects in spermatogonial differentiation, with absence of mature sperm.

Simultaneous knockdown of mei-P26dsRNA.shRNA.Scer\UAS.P\T and U2AdsRNA.shRNA.Scer\UAS.P\T driven by Scer\GAL4bam.T:Hsim\VP16 causes a more severe phenotype compared with individual knockdown, with increased accumulation of spermatogonial cysts and fewer spermatocytes in the apical region of the testes.
