Dis31/Dis31 mutants survive at the second instar larval stage for an extended period and then die. Dis31/Dis31 wing disc clones in second instar larvae have significantly fewer cells per clone compared to controls and mutant cells undergo only one cell division in a 48hr time window (compared to 4 in wild type cells).

Dis31/Dis32 third instar larvae with expression of Dis3S786E.Scer\UAS.P\T driven by Scer\GAL4tub.PU lack imaginal discs, have reduced brain and ventral nerve cord size, whereas other tissues that do not grow via mitotic cell cycles (salivary glands) are generally normal in size and morphology; larval neuroblasts have multipolar spindles; larval neuroblasts have multipolar, disorganized or short spindles, 5% of mitotic neuroblasts display aneuploidy, 44% display over condensed chromosomes and more neuroblasts are mitotically active than controls.

Dis31/Dis32 larvae have shortened mitotic spindles in neuroblasts.

Dis3G5039/Dis31 flies are viable, largely infertile and morphologically normal (apart from small bristles and occasional crumpled wings); a large percentage of stage 5-6 egg chambers show over proliferation, with excess posterior follicle cells.

Comments

Expression of Dis3S786E.Scer\UAS.P\T driven by Scer\GAL4tub.PU rescues larval development until late wandering third instar larval stage (though larvae take 7-12 extra days to reach this stage and then die shortly after) in Dis31/Dis31 mutants.

Expression of Dis3G5039 driven by Scer\GAL4Act.PU rescues over proliferation of follicle cell phenotypes in Dis3G5039/Dis31 egg chambers.
