Description

UAS regulatory sequences drive expression of Bant\lef coding sequences where the bacterial signal sequences, comprising the first 33 amino acid residues have been removed and are replaced with a methionine preceded by an optimal Drosophila Kozak's sequence (GCCAGC).

Expression of Zzzz\lefScer\UAS.cGa in the wing disc primordium under the control of Scer\GAL4bs-1348 produces no obvious phenotype.

Expression of Zzzz\lefScer\UAS.cGa in the wing disc primordium under the control of Scer\GAL4Bx-MS1096 produces no obvious phenotype.

Expression of Zzzz\lefScer\UAS.cGa under the control of Scer\GAL4tsh-md621, Scer\GAL4twi.PG or Scer\GAL4Act.PU results in almost complete lethality.

Expression of Zzzz\lefScer\UAS.cGa under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 results in near complete lethality. Embryos exhibit a range of developmental defects. The least severely affected embryos have a U-shaped phenotype, which is typical of mutations interfering with dorsal closure. More severely affected embryos are greatly reduced in size, have dorsal holes in the cuticle, and lack all head skeletal structures. In addition, these embryos exhibit severe defects in the F-actin network in leading-edge cells, which forms a regular line in wild-type embryos.

Expression of Zzzz\lefScer\UAS.cGa under the control of Scer\GAL4pnr-MD237 results in a dorsal cleft phenotype.

Strong expression of Zzzz\lefScer\UAS.cGa in the wing disc under the control of Scer\GAL4Bx-MS1096 causes small scooped wings, which are elongated along the proximal-distal axis and distal vein truncations. Males with three copies of the Zzzz\lefScer\UAS.cGa transgene, driven by Scer\GAL4Bx-MS1096 at 29[o]C exhibit the small wing vein phenotype with almost 100% penetrance, approximately 50% of these individuals also exhibit vein truncations.

Co-expression of Zzzz\EFScer\UAS.cGa and Zzzz\lefScer\UAS.cGa in the wing disc primordium under the control of Scer\GAL4bs-1348 produces a strong and penetrant wing margin notching phenotype.

Co-expression of Zzzz\EFScer\UAS.cGa and Zzzz\lefScer\UAS.cGa in the wing disc primordium under the control of Scer\GAL4Bx-MS1096 produces a strong and penetrant wing margin notching phenotype.

Co-expression of Rab11S25N.Scer\UAS.P\T.T:Avic\GFP-YFP with Zzzz\lefScer\UAS.cGa in the wing disc primordium produces a strong and penetrant wing margin notching phenotype (when both are under the control of Scer\GAL4Bx-MS1096).

Moderate expression of Bant\lefUAS.cGa under the control of Scer\GAL4Bx-MS1096 in rhove-1 heterozygotes results in wing vein loss, while moderate Bant\lefUAS.cGa expression (with Scer\GAL4Bx-MS1096) alone doesn't have this phenotype.

Moderate expression of Bant\lefUAS.cGa under the control of Scer\GAL4Bx-MS1096 in vnunspecified heterozygotes results in wing vein loss, while moderate Bant\lefUAS.cGa expression (with Scer\GAL4Bx-MS1096) alone doesn't have this phenotype.

Moderate expression of Bant\lefUAS.cGa under the control of Scer\GAL4Bx-MS1096 in kniri-1 heterozygotes results in wing vein loss, while moderate Bant\lefUAS.cGa expression (with Scer\GAL4Bx-MS1096) alone doesn't have this phenotype.

Expression of Bant\lefUAS.cGa does not suppress the ectopic vein phenotype of rlSem.cUa.UAS under the control of Scer\GAL4Bx-MS1096.

Expression of Bant\lefUAS.cGa suppresses the strong ectopic vein phenotype of RafAct.UAS under the control of Scer\GAL4CY2.

Moderate expression of Bant\lefUAS.cGa under the control of Scer\GAL4Bx-MS1096 in Dsor1r1 results in an aggravated Bant\lefUAS.cGa (Scer\GAL4Bx-MS1096) phenotype consisting of more pronounced scooped wings and distal vein truncations.
