Deletion in the first exon of the dnc[RA] transcript (extending from -2148 to +71 relative to the transcription start site of dnc[RA]). This reduces the level of the dnc[RA] transcript almost to zero, slightly reduces dnc[RL] expression and does not affect the expression of the other dnc transcript groups.
The reduced heat-induced ethanol (EtOH) resistance characteristic for hangAE10 mutant adults is not exacerbated further by combination with dncΔ143 which on its own also lead to a decrease in the heat > ethanol cross-tolerance.
hangAE10;dncΔ143 double mutant adult flies display normal ethanol tolerance development upon re-exposition to EtOH, unlike each of the single mutants that show significantly reduced tolerance buildout.
The reduced ability of dncΔ143 mutant adults to develop tolerance to ethanol upon re-exposition is rescued by expression of dncScer\UAS.all when driven by either Scer\GAL4dnc.RA (alone or combined with Scer\GAL80ple.PK to restrict the expression pattern) or Scer\GAL4NP6510 but not when driven by Scer\GAL4GMR58E02. The tolerance buildout can also be rescued by Scer\GAL4dnc.RA-driven expression of dncScer\UAS.RA.T:Avic\GFP-EGFP but not dncScer\UAS.RG.T:Avic\GFP-EGFP or dncScer\UAS.RL.T:Avic\GFP-EGFP.
