The co-expression of HPV18\E6UAS.Tag:MYC and Hsap\UBE3AUAS.cRa under the control of Scer\GAL4ap.PU in a Magibst-homozygous background leads to pupal lethality.

The co-expression at room temperature of HPV18\E6UAS.Tag:MYC and Hsap\UBE3AUAS.cRa under the control of Scer\GAL4ap.PU leads to fully penetrant adult wing defects, including smaller and blistered held-out wings that are full of melanized tissue, while co-expression at 29[o]C leads to increased apoptosis in the dorsal compartment of third instar larval wing discs (assessed by the number of Casp3-positive cells), as compared to controls; the severity of adult wing defects is ameliorated by additional co-expression of MagiUAS.mCherry or MagiΔWW.UAS.Tag:FLAG, but not of MagiΔPDZ.UAS.Tag:FLAG, scribUASp.GFP or dlg1UAS.EGFP, while the third instar larval wing disc defects are rescued by the additional co-expression of MagiUAS.mCherry but not of scribUASp.GFP or bskDN.UAS; additional co-expression of BacA\p35UAS.cHa also rescues the increased apoptosis at the dorsal compartment of third instar larval wing discs without leading to disc overgrowth, but induces basal delamination of cell clusters, as compared to controls.

The triple co-expression of HPV18\E6UAS.Tag:MYC, Hsap\UBE3AUAS.cRa and Ras85DV12.cUa.UAS under the control of Scer\GAL4ap.PU from the first or second instar larval stage onwards (by the use of Gal80[ts]) results in the most highly expressing cells within the dorsal compartment of the third instar larval imaginal disc adopting a fibroblast-like and flat morphology, displaying filopodial-like processes and delaminating at the basal side of the columnar epithelium, although without crossing the dorsal-ventral boundary, as compared to controls.

The co-expression of HPV18\E6UAS.Tag:MYC and Hsap\UBE3AUAS.cRa under the control of Scer\GAL4GMR.PU leads to a fully penetrant rough eye phenotype, disorganized and fused ommatidia (resulting in increased numbers of cone cells, bristle cells and primary, secondary and tertiary pigment cells per ommatidium), and abnormal adherens junctions between photoreceptors, as compared to the single expressions or wild-type controls; this rough eye phenotype is enhanced by the co-expression of either InRUAS.cUa, leading to overgrowth and out-pocketing of the eye tissue, or InRDN.UAS.cUa, leading to severe eye necrosis, but is not modified by heterozygosity for Akt104226, hhAC, witA12, Tak12527, BuffyH37, DebclE26, hop2 or Stat92E06346, or by the additional co-expression of rlSem.cUa.UAS, armS10.UAS.Tag:MYC, ykiUAS.EGFP,Tag:HA, bskDN.UAS, DebclUAS.cUa or p53H159N.UAS.Ex.

The triple co-expression of HPV18\E6UAS.Tag:MYC, Hsap\UBE3AUAS.cRa and either Ras85DV12.cUa.UAS or Nint.U.UAS under the control of Scer\GAL4GMR.PU leads to the migration of a considerable proportion of expressing eye tissue cells into distant pupal tissues (e.g. abdomen), as compared to controls; the Nint.U.UAS triple co-expression condition also leads to pharate lethality, while the Ras85DV12.cUa.UAS triple co-expression condition does not suppress the pupal lethality induced by the single expression of Ras85DV12.cUa.UAS.
