Inhibition (using Ctet\TeTxLCTNT.UAS) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT002081 and Hsap\RELAAD.R23E07 in third instar larvae significantly reduces the ability of larvae to navigate to an appetitive odor, compared to controls; there is no difference in run speed, compared to controls; turn rate is significantly reduced when larvae are moving down, but not up, an odor gradient, and not in the absence of odor, compared to controls; the probability of turning towards, rather an away from, an appetitive odor source is not affected, compared to controls.

Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 625 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT002081 and Hsap\RELAAD.R23E07 in third instar larvae significantly increases stopping and head-casting behavior, compared to controls; the stopping phenotype is not affected by the odor environment, whereas the head casting phenotype is more likely and more vigorous when larvae are moving down, rather than up, an odor gradient.

Unilateral single-cell optogenetic activation (using Cnoc\ChR1Cs.20xUAS.FRT.Venus and the low activity flippase Scer\FLP1G5.R57C10.Tag:PEST(mOdc), activated with 625 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT002081 and Hsap\RELAAD.R23E07 in third instar larvae is sufficient to induce stopping and head-casting behaviors, relative to baseline.
