Kdm5140 homozygotes show a slow-growth phenotype that results in larvae taking longer to develop to the same size as controls animals. The third instar larval muscle exhibits altered NMJ morphology, with decreased number and increased size of type Ib synaptic boutons, which present unbundled microtubules.

In Kdm5140 homozygous third instar larvae bearing two copies of Kdm5JmjC.Tag:HA, muscles exhibit altered NMJ morphology and physiology, with decreased number of synaptic boutons and decreased eEJC amplitudes. These larvae show decreased crawling ability (decreased crawling length, decreased average and maximum crawling speed, and decreased speed normalized to body length).

In Kdm5140 homozygous third instar larvae bearing two copies of Kdm5L854F.Tag:HA, muscles exhibit altered NMJ morphology, with decreased number and increased size of type Ib synaptic boutons, which present unbundled microtubules. These larvae show decreased crawling ability (decreased crawling length, decreased average and maximum crawling speed, and decreased speed normalized to body length).

Kdm5140 homozygotes exhibit developmental delay compared to controls.

Kdm5140 wandering third instar larvae exhibit significantly decreased number of dividing cells (PH3+) and significantly increased cell death (Dcp-1+) in the wing pouches compared to controls.

Kdm5140 third instar larvae exhibit a significant decrease in the number of prothoracic gland cells undergoing S phase of endocycling compared to controls.

lid140 mutants are lethal; pharate adults have morphologically normal heads, thoraces, legs, and abdominal segmentation, but fail to eclose.

lid140 homozygotes show a severe delay to pupariation, a significant decrease in larval weight, but do not show defects in food intake, as compared to controls. Despite taking 5 days longer, the final larval, imaginal disc, and pupal size are normal; the pupal wing shows similar growth of the axon along the L3 wing vein, in addition to the correct number and position of the sensory bristles along the anterior margin of the wing, as compared to controls.

Larval wings disc are significantly smaller at 5.5 after egg laying, exhibit a significant decrease in cell division (mitotic index/phospho-H3) and exhibit a significant increase in apoptosis (Dcp-1-positive cells). Upon irradiation, the wing discs show increased apoptosis, as compared to irradiated controls (Dcp-1 immunostaining levels).

Expression of Kdm5UASp.cSa under the control of Scer\GAL432B, Scer\GAL4Mai60, Scer\GAL4spok.1.45 or Scer\GAL4phm.PO in the Kdm5140 homozygous background leads to curved-down wings compared to controls.
