UAS regulatory sequences drive expression of an N-terminally deleted form of Akt1; the first 341 bp of the coding sequence of Akt1-RA are missing, resulting in a deletion of 75% of the PH domain in the encoded protein. The coding sequence is tagged with a myristoylation signal.

Expression of AktΔPH.UAS.Tag:Myr(Unk) under the control of Scer\GAL4en-e16E leads to an increase in cell death in posterior compartment of third instar larval wing discs.

Expression of Akt1ΔPH.UAS.Tag:Myr(Unk) under the control of Scer\GAL4Tub.PU in midgut MARCM clones leads to a significant decrease in cell number but an increase in cell size when compared to control adults.

Expression of Akt1ΔPH.UAS.Tag:Myr(Unk) under the control of Scer\GAL4nSyb.PS does not lead to any synaptic overgrowth at neuromuscular junction (assessed by number of boutons and NMJ length) nor any significant changes in crawling velocity in third instar larvae.

Expression of Akt1ΔPH.UAS.Tag:Myr(Unk) under the control of Scer\GAL4tim.UAS, dramatically increases sensitivity to oxidative stress. These flies die within 2 days of being challenged with 1mM paraquat. When treated with 0.05-0.1mM paraquat they survive up to 6 days, with the majority showing arrhythmic locomotor behavior.

Expression of Akt1ΔPH.UAS.Tag:Myr(Unk) in the fat body, under the control of Scer\GAL4Lk6-DJ634, increases sensitivity of behavioral rhythms to paraquat.
