    Detailed Description
    Statement
    Reference

Eaat1hypo homozygous larvae have severely impaired locomotion and their neuromuscular junctions exhibit morphological and functional defects compared to controls: there are more boutons per muscle but boutons (type Ib) are smaller and there are no changes in the number of active zones per muscle; there are significant increases in EJP amplitude, quantal content and burst duration, but no change in mEJP amplitude; there are no changes in muscle area. Eaat1hypo homozygous larvae do not show any obvious VNC structural defects, exhibiting similar numbers of neurons and astrocytes to controls.

Eaat1hypo/Eaat1SM2 transheterozygous larvae have severely impaired locomotion and their neuromuscular junctions exhibit morphological and functional defects compared to controls, namely a significant increase in burst duration and a decrease in burst frequency, significantly more but smaller boutons (including satellite boutons) and smaller muscles.

    Comments

Constitutive expression of Eaat1UAS.Venus under the control of Scer\GAL4repo rescues the locomotor defects of Eaat1hypo homozygous larvae; the same rescue is obtained by conditional expression only during L1-L3 , L2-L3, L3, or mL3 (temporal regulation of expression is under the control of Gal80[ts] and temperature shift from 18[o]C to 29[o]C).
