Expression of Scer\ABP140Scer\UAS.P\T.T:Avic\GFP-EGFP in the somatic follicle cells under the control of Scer\GAL4c355 has little or no effect on female fertility, and no apparent defects in F-actin are seen at stage 9 or 10B of follicle development. Similar results are seen when Scer\ABP140Scer\UAS.P\T.T:Avic\GFP-EGFP is expressed using the weak germline driver Scer\GAL4nos.UTR.T:Hsim\VP16, except that mild defects in cortical actin integrity and actin bundle organisation are observed at stage 10B. Conversely strong germline expression under the control of either Scer\GAL4mat.αTub67C.T:Hsim\VP16 or Scer\GAL4osk.T:Hsim\VP16 results in female sterility. At stage 9 of follicle development, severe defects in F-actin and follicle morphology are seen, including the loss of cortical actin integrity, which results in aggregates of ring canals and multi-nucleate nurse cells, altered nurse cell and oocyte shape and the formation of phalloidin and Scer\ABP140Scer\UAS.P\T.T:Avic\GFP-EGFP threads in the germinal vesicle. At stage 10B cortical actin is compromised, as multinucleate nurse cells are always observed. At 25[o]C, often all 15 nurse cell nuclei are contained within one or two "cells". Aberrant F-actin structures are also observed around the germinal vesicle.
