Summaries
Gene Group (FlyBase)
ACTINS -
Actins are highly conserved eukaryotic filament-forming proteins that are major components of the cytoskeleton. Actin polymerisation is controlled by ATP binding and hydrolysis. Actins are involved in various cell processes including cell movement, muscle contraction, cell trafficking and mechanical support. (Adapted from PMID:21859859).
TIP60 COMPLEX -
The Tip60 chromatin remodeling complex acetylates nucleosomal phospho-histone H2Av and exchanges it with unmodified H2Av. (Adapted from FBrf0185742).
Protein Function (UniProtKB)
Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. Multiple isoforms are involved in various cellular functions such as cytoskeleton structure, cell mobility, chromosome movement and muscle contraction.
(UniProt, P10981)
Phenotypic Description (Red Book; Lindsley and Zimm 1992)
Act87E
Encodes actin found in larval muscle and adult cephalic and abdominal muscle (Fyrberg, Mahaffey, Bond, and Davidson, 1983, Cell 33: 115-23).

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry P10981)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

Subunit Structure (UniProtKB)

Component of the Tip60 chromatin-remodeling complex which contains the catalytic subunit Tip60 and the subunits Domino, Tra1, Brd8, E(Pc), DMAP1, Pontin, Reptin, Ing3, Act87E, BAP55, Mrg15, MrgBP, Gas41 and YL-1.

(UniProt, P10981)
Post Translational Modification

N-terminal cleavage of acetylated cysteine of immature actin by ACTMAP.

Oxidation of Met-45 by Mical to form methionine sulfoxide promotes actin filament depolymerization. Methionine sulfoxide is produced stereospecifically, but it is not known whether the (S)-S-oxide or the (R)-S-oxide is produced.

Testis-specificity index

The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).

Additional Descriptive Data

Act87E expression is observed in the adult dorsal vessel but not in the larval dorsal vessel.

Act87E transcripts are most abundant in larvae. They accumulate in a small subset of larval segmentally repeated muscles. The level of transcript accumulation is much lower than that of Act57B transcripts.

ChEST reveals this is a target of Mef2.

In a sample of 79 genes with multiple introns, 33 showed significant heterogeneity in G+C content among introns of the same gene and significant positive correspondence between the intron and the third codon position G+C content within genes. These results are consistent with selection adding against preferred codons at the start of genes.

Age related changes in Act87E expression have been studied by Northern analysis.

Sequence analysis demonstrates insect muscle actins form a distinct family of closely related proteins significantly diverged from cytoplasmic actins.

Translation of actin RNA in early embryos injected with initiation factors has been studied.

No lethal mutations in Act87E gene were isolated during a molecular and genetic analysis of region including Act87E gene. Possible explanations include non-lethality of Act87E mutations or substitution of Act87E function by other actin-encoding genes in the genome.

Encodes actin found in larval muscle and adult cephalic and abdominal muscle (FBrf0038990).

              Additional comments

The Act87E gene may have been derived from the Act5C gene by retroposition.

The Act87E gene may have been derived from the Act57B gene by retroposition.
