Phenotypic Description (Red Book; Lindsley and Zimm 1992)
Aprt: Adenine phosphoribosyltransferase
Is the structural gene for adenine phosphoribosyl-transferase [APRT, AMP: pyrophosphate phosphoribosyl-transferase (EC 2.4.2.7)], a homodimer with 23,000 dalton subunits. It is a purine salvage enzyme which catalyzes the synthesis of AMP from 5-phosphoribosyl-1-pyrophosphate. Flies homozygous for a null allele Aprt1 survive on 15 times the concentration of purine that wild type tolerates and show about 2% wild-type enzyme activity; Aprt1/+ exhibit about half wild-type activity. Aprt2 has 9% normal enzyme activity. The dosage response suggests that the mutant affects the structural gene for APRT.

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry P12426)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

      Other Comments

DNA sequence conservation between Dpse\Aprt and Aprt has identified a nuclear matrix attachment site. A palindrome with direct repeats of non-conserved DNA sequences may act as a promoter for Aprt.

Comparison of CpG distribution in the coding region of 121 genes from six species supports the mCpG mutational hotspot explanation of CpG suppression in methylated species at position II-III and III-I.

In Aprt and Dpse\Aprt the use of alternate 5' splice sites to process an intron in pre-mRNA leads to simultaneous production of RNAs encoding nonfunctional proteins in addition to functional Aprt or Dpse\Aprt, respectively.

The structural gene for adenine phosphoribosyltransferase.

Mutant flies are resistant to the toxic effects of dietary purine.
