Expression studied both in vitro and in vivo. Imp-E denotes
three "early" genes involved in elongation of appendage-forming regions of the imaginal discs by means of epithelial-cell rearrangements (Natzle et al., 1988).

Northern blotting reveals that the highest levels of ImpE2 transcript are present in white prepupae and early (24hr) pupae. Moderate levels are seen in 12-18 hr embryos and in late pupae (>48 hr). Low levels of transcript are found in 3-12 hour embryos and in larvae, while no transcripts are detected in adults, or in 0-3 hour embryos. When third instar larval imaginal discs are cultured for 3, 6, 9 or 12 hr in the presence of 20-hydroxyecdysone (20-HE), the 1.7 kb ImpE2 transcript is detected, while no transcript is detected in discs cultured for the same length of time without 20-HE. Radioactive in situ hybridization shows that in 12-15 hour embryos, transcript localizes to the ectoderm. In 0-4 hour prepupae, part of the columnar epithelium and the peripodal epithelium of each imaginal disc, and the epithelium of the proventriculus, accumulates ImpE2 transcript.
After 12 hours of embryogenesis but prior to dorsal closure, the ImpE2 protein is localized cytoplasmically in a punctate pattern, whereas after dorsal closure, it is found in an extracellular layer adjacent to the apical ectodermal surface. Embryos between these stages show a combination of staining, with punctate cytoplasmic (possibly vesicular) staining posteriorly, and extracellular staining anteriorly. ImpE2 protein is likely secreted apically. E.M. immunolocalization to prepupal imaginal discs shows ImpE2 protein to be localized to vesicles at (and fusing to) the apical cytoplasm. Some staining is also seen at the cell surface and the extracellular matrix.
