In situ analysis of male adults shows that expression of PpY-55A transcript is testis-specific.

PpY-55A transcripts are detected in males, pupae and at a low level in larvae. They are detected in testes in larvae and males but not in the carcasses from which the gonads were removed.

PpY-55A protein is detected only in extracts from testis on western blots. Immunolocalization studies demonstrate that it is found predominantly in the nuclei of cells distributed along the length of the testis. Staining is weak in the apical region. Colocalization studies with an enhancer trap marker line show that the site of PpY-55A protein expression is the cyst cells.

        dsRNA made from templates generated with primers directed against this gene tested in RNAi screen for effects on Kc167 and S2R+ cell morphology.

        PpY-55A is a testis specific protein phosphatase with biochemical properties distinct from rabbit PP1. Localisation of PpY-55A to somatic cyst cells in testis indicates the protein may be important for differentiation of mature spermatozoa.

        Isolated from a Drosophila head cDNA library, using a rabbit serine/threonine-specific protein phosphatase (PP-1) cDNA fragment as a probe.
