Gene Group (FlyBase)
WNTs -
WNTs are evolutionarily conserved secreted Cys-rich glycoproteins, defined by sequence homology to the original members of the family - Wnt1 in mouse and wingless (wg) in Drosophila. They are extracellular ligands for members of the Frizzled family of receptors as well as other receptors. (Adapted from PMID:23151663).
Pathway (FlyBase)
Wnt-TCF Signaling Pathway Core Components -
The canonical Wnt signaling pathway is initiated by the binding of a Wnt ligand to a frizzled family receptor on the cell surface. Activation of the pathway leads to the inhibition of cytoplasmic β-catenin (arm) degradation and its subsequent accumulation in the nucleus, where it regulates the transcription of target genes. (Adapted from FBrf0218499 and FBrf0223299).
Protein Function (UniProtKB)
Binds as a ligand to a family of frizzled seven-transmembrane receptors and acts through a cascade of genes on the nucleus. Segment polarity protein. May function in gonadogenesis and limb development. Wg and Wnt2 have a role in the developing trachea and together are responsible for all dorsal trunk formation.
(UniProt, P28465)
Summary (Interactive Fly)

ligand - Wnt homolog - required for the development of the sheath of the male reproductive tract and testis morphogenesis - mediates redox regulation - maintains the germ line stem cell differentiation niche

Post Translational Modification

Palmitoleoylated by porcupine. The lipid group functions as a sorting signal, targeting the ligand to polarized vesicles that transport Wnt2 to unique sites at the cell surface. Depalmitoleoylated by notum, leading to inhibit Wnt signaling pathway.

Additional Descriptive Data

Wnt2 transcript is first detected in stage 13 embryos, in the mesodermal cells which will form the somatic mesoderm of the gonad. Wnt2 is expressed in the gonad at stage 15, and this expression is limited to the posterior portion of the gonad at stage 16. In late third instar larvae, Wnt2 expression is detected in male genital primordia. In the 24 hr male pupal genital disc, expression is detected in the epithelial cells at the apical tip of the developing seminal vesicle. Wnt2 transcript is not detected in female structures; however, when Wnt2 is ectopically expressed in third instar larval females, male-specific pigment cells are formed in the resulting adults.

Wnt2 promotes correct patterning of direct flight muscles through a mechanism that is independent of the attachment site differentiation initiated by sr.

Wnt2 assists wg to specify the main tracheal trunk. Wnt2 affects tracheal development but apperently has no effect on the cuticle, whereas wg influences both.

Candidate gene for testis length quantitative trait locus.

Wnt2 is required for cell fate determination and morphogenesis in the developing male reproductive tract.

Wnt2 encodes an amino terminal signal peptide suggesting the gene product is a secreted protein.

Sequence analysis of Wnt genes is performed in several species to determine the ancestral lineage of the gene family.
