Gene Group (FlyBase)
C2H2 ZINC FINGER TRANSCRIPTION FACTORS -
Zinc finger C2H2 transcription factors are sequence-specific DNA binding proteins that regulate transcription. They possess DNA-binding domains that are formed from repeated Cys2His2 zinc finger motifs. (Adapted from PMID:1835093, FBrf0220103 and FBrf0155739).
Protein Function (UniProtKB)
May belong to a complex set of multifingered proteins which play an important role in gene activation or regulation at early embryonic stages through a maximal accumulation of their transcripts (or protein product) in the mature oocyte.
(UniProt, P15619)
Phenotypic Description (Red Book; Lindsley and Zimm 1992)
Sryc: Serendipity cognate
Structural gene for a protein of 868 amino acids with eight TFIIIA-like fingers. Developmental Northern blots reveal abundant transcript in adult females but not males and during the first four hours of embryonic development; low levels present in later embryos, larvae, and pupae. In situ hybridization to tissue sections shows that transcripts produced in nurse cells and beginning at stage 10 are transported to and accumulate in oocytes.
*wdn: wings down
Wings extended and drooping or even directed ventrally, broad with close crossveins. Overlaps wild type. Low viability. RK3.

Additional Descriptive Data

wdn transcripts are most abundant in early embryos and virgin females on northern blots but are detected in all stages tested. They are detected by in situ hybridization in nurse cells up to stage S10A and then are transferred to the oocyte. Transcript levels decline after egg deposition and before the cellular blastoderm stage. Transcripts are uniformly distributed during gastrulation but not at later stages.

Additional Descriptive Data

wdn protein is detected in ovaries in the nucleus of nurse cells in stages S1-S6. In stages S7 and S8 it is no longer detectable in nurse cells and is observed in one or two densely stained spherical objects in the oocyte nucleus. In stages S9 and S10 staining is again observed in nurse cell nuclei. The staining in the spheres in the oocyte nuclei gradually fades during stage 9 and the staining becomes uniform over the entire oocyte nucleus and persists in later stages of oogenesis. wdn protein is detected at a lower level in the nuc ei of border cells and follicle cells. Low levels of wdn were oberved in embryos and larvae.

In a sample of 79 genes with multiple introns, 33 showed significant heterogeneity in G+C content among introns of the same gene and significant positive correspondence between the intron and the third codon position G+C content within genes. These results are consistent with selection adding against preferred codons at the start of genes.

Functional dissection of the promoter using Ecol\lacZ reporter constructs identifies the minimal promoter sequences between position -210 and +30, relative to the transcription start site. These sequences subdivide into follicle cell- and germ line specific regions and appear to function in a direction-independent and distance-sensitive manner.

wdn protein expression has been analysed.

wdn zinc finger protein product is present in the oocyte nucleus and during embryonic and larval development. Homozygous mutants are zygotic lethal and display a disorganization of the bodyplan.
