Helicase at 25E (Hel25E) encodes the UAP56 RNA helicase in the DECD subfamily of DEAD-box helicases. It is an enhancer of position-effect variegation, and contributes to mRNA splicing, nuclear export, and piRNA biosynthesis. The product of Hel25E binds heterochromatic cluster transcripts, along with the product of vas, to organize a piRNA-processing compartment that spans the nuclear envelope to silence transposon transcripts. TRANSCRIPTION EXPORT COMPLEX -
The Transcription and export (TREX) complex is a multisubunit complex, which acts throughout RNA biogenesis to couple transcription with nuclear export. (Adapted from FBrf0233979 and PMID:27679854).
Protein Function (UniProtKB)
Required for mRNA export out of the nucleus. Probable RNA helicase that may regulate entry into mitosis by down-regulating the expression of other genes whose activity may be rate-limiting for entry into mitosis during embryogenesis. Binds to salivary gland chromosomes and modifies position effect variegation. Promotes an open chromatin structure that favors transcription during development by regulating the spread of heterochromatin.

Additional Descriptive Data
In the larval salivary gland, Hel25E protein associates with the polytene chromosomes. In ovaries, staining is seen in nurse cell and follicle cell nuclei. Confocal microscopy of cycle 14 embyros double-labeled with antibodies against Hel25E and betaTub60D shows that Hel25E disappears from within mitotic domains, although it is present in the nuclei of the other cells at the surface of the embryo.

    RNAi screen using dsRNA made from templates generated with primers directed against this gene results in chromosome misalignment on the metaphase spindle when assayed in S2 cells. This phenotype can be observed when the screen is performed with or without Cdc27 dsRNA.
    Identified as a potential component of the hh signalling pathway in a genome-wide RNAi screen. dsRNA made from templates generated with primers directed affects the extent of expression of a hh signaling reporter construct in Clone 8 cells.
    Hel25E encodes a putative RNA helicase that contains an unusual variant (DECD) of the DEAD motif common to these proteins.
    Deletion or mutation of the Hel25E locus enhances position effect variegation of w.
    Hel25E gene can influence position effect variegation (PEV). Loss of one or two copies enhances PEV of a variegating allele of the w gene, complete removal of Hel25E is lethal.
    S.pombe Spom\wee1 Spom\mik1 double mutants are functionally complemented by Hel25E.
