Hormone-receptor-like in 78 (Hr78) encodes a protein induced above a basal level by 20-hydroxyecdysone. Hr78 mutants die during mid-larval stages with defects in molting of the tracheal cuticle. [Date last reviewed: 2019-03-07]

Protein Family (UniProt)
Belongs to the nuclear hormone receptor family. NR2 subfamily. (Q24142)
Summaries
Gene Group (FlyBase)
NUCLEAR RECEPTOR (LIGAND-DEPENDENT) TRANSCRIPTION FACTORS -
Nuclear receptors (NR) are C4 zinc finger ligand-dependent DNA-binding transcription factors. Members of the NR superfamily are defined by the presence of a highly conserved DNA-binding domain and a less conserved C-terminal ligand-binding domain. (Adapted from FBrf0184203).
Protein Function (UniProtKB)
Binds to direct repeats of the sequence 5'-AGGTCA-3'. Inhibits the ecdysone response. Plays an essential role in regulating molting of the tracheal cuticle during larval development.
(UniProt, Q24142)

Additional Descriptive Data

Hr78 is expressed throughout third instar larval and prepupal development. A distinct increase in levels is seen at 106hr after egg laying suggesting that it may be ecdysone-inducible. Hr78 transcripts were shown to be ecdysone inducible in cultured larval organs.

Hr78 transcripts are expressed at low levels in embryos and early larvae and increase after the 2nd to 3rd instar molt. Expression does not appear to be strongly activated by ecdysone. They are expressed ubiquitously in embryos and imaginal discs at low levels.

    Additional comments

FlyBase curator comment: the insertion in the "e03593" Exelixis line (PBac{RB}CG33214e03593) was originally assigned to the Hr78 gene in FBrf0179797, resulting in the "Hr78e03593" (FBal0160320) allele. However, FBrf0184340 shows that the insertion is actually within CG33214.

    Other Comments

Hr78 protein is an obligate partner for Samuel protein, a cofactor that requires Hr78 protein for its stability.

Hr78 is not required for morphogenetic furrow formation in the developing eye.

Hr78 functions at the top of the ecdysteroid regulatory hierarchies. Results propose that Hr78 is a ligand-activated receptor that plays a central role in directing the onset of metamorphosis.

Null mutations lead to lethality during the third larval instar with defects in ecdysteroid-triggered developmental responses. Mutants fail to activate the mid-third instar regulatory hierarchy that prepares the fly for metamorphosis.

Hr78 may play a regulatory role in the transcriptional cascade triggered by the hormone ecdysone.
