Gene Snapshot
Retinoblastoma-family protein (Rbf) binds to members of the E2F transcription factors and forms the DREAM complex, which regulates genes with diverse biological functions including cell cycle regulators, apoptosis regulators, and DNA replication/repair factors. Rbf can also bind chromosomal proteins and modulate genome stability. Rbf is negatively regulated by CycE-Cdk2 and CycD-Cdk4 cyclin-dependent kinase complexes. [Date last reviewed: 2016-12-01]

Summaries
UniProt Contributed Function Data
Functions in cell cycle regulation. Component of the DREAM complex, a multiprotein complex that can both act as a transcription activator or repressor depending on the context. In follicle cells, the complex plays a central role in the site-specific DNA replication at the chorion loci. During development, the complex represses transcription of developmentally controlled E2F target genes.

Subunit Structure (UniProtKB)
Forms a complex with the DRTF1/E2F transcription factor through binding to a C-terminal region of E2F. This binding inhibits the E2F-mediated transactivation activity. Component of the DREAM complex at least composed of Myb, Caf1, mip40, mip120, mip130, E2f2, Dp, Rbf, Rbf2, lin-52, Rpd3 and l(3)mbt.
(UniProt, Q24472)
Post Translational Modification
Phosphorylation by cyclin E-dependent kinases appears to negatively regulate RBF activity.

gig and Rbf synergistically induce oxidative stress via increased protein synthesis, inhibited de novo lipid synthesis, and decreased reactive oxygen species enzyme induction.
dsRNA made from templates generated with primers directed against this gene tested in RNAi screen for effects on Kc167 and S2R+ cell morphology.
E2f2 as part of the E2f-Rb complexes has a role in the down regulation of CycE during the gap phase of endocycling cells in normal development.
Rbf is required at multiple stages of development. Most, if not all, of the function of Rbf during development is mediated through E2f.
Rbf is dispensable for early cell cycles in the embryo, but plays an essential role in the introduction of G1 control during development.
Identification: Yeast two hybrid screen for proteins that interact with E2f.
Rbf plays a critical role in the regulation of cell proliferation.
Overexpression of dap during eye development interferes with cell cycle progression and interacts genetically with Rbf and CycE.
