Cbl protein lacks proline-rich sequences at the carboxy-terminus that are found in related proteins which allow binding to SH3-domain containing adaptor proteins. An in vitro protein interaction assay was used to show that Cbl protein does not interact with drk protein. It does, however associate with activated Egfr protein in vivo.

Both Cbl protein isoforms, as assayed by Western blot analysis, are expressed at all embryonic stages, all larval instars, and in ovaries. The short isoform is predominant at all stages.
