GO Summary Ribbons
Gene Group (FlyBase)
Protein Family (UniProt)
Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. PDPK1 subfamily. (Q9W0V1)
Summaries
Gene Group (FlyBase)
AGC KINASES (UNCLASSIFIED) -
The AGC kinase subfamily are serine/threonine kinases that were originally defined based on the sequence similarity of the catalytic domain. The AGC KINASE (UNCLASSIFIED) gene group represents AGC kinases that do not fall into the PKA, PKC or PKG families. (Adapted from FBrf0132098 and PMID:20027184).
Pathway (FlyBase)
Insulin-like Receptor Signaling Pathway Core Components -
The Insulin-like Receptor (IR) signaling pathway in Drosophila is initiated by the binding of an insulin-like peptides to the Insulin-like receptor (InR). (Adapted from FBrf0232297, FBrf0230017 and FBrf0229989.)
Protein Function (UniProtKB)
Serine/threonine kinase required for embryonic development. Inhibits apoptosis. Acts in the insulin receptor transduction pathway which regulates cell growth and organ size, by phosphorylating and activating Akt1 and S6k. May be involved in axonal pathfinding and synaptogenesis, and in spermatogenesis.
(UniProt, Q9W0V1)
Summary (Interactive Fly)
a master kinase that is crucial for the activation of AKT (PKB) and many kinases including PKC and S6K - contains pleckstrin homology (PH) domains that interact with the PtdIns(3,4,5)P second messenger - regulates cell growth - controls synaptic bouton size, active zone number, and synaptic function - essential for neuronal differentiation

Domain
The PIF-pocket is a small lobe in the catalytic domain required by the enzyme for the binding to the hydrophobic motif of its substrates. It is an allosteric regulatory site that can accommodate small compounds acting as allosteric inhibitors.

dsRNA made from templates generated with primers directed against this gene used to treat S2 cells.
dsRNA made from templates generated with primers directed against this gene inhibits TORC1-dependent S6k phosphorylation but does not inhibit Akt1 phosphorylation in S2 cells. In fact, knockdown of Pk61C increases TORC2-dependent Akt1 phosphorylation.
When dsRNA constructs are made and transiently transfected into S2 cells in RNAi experiments, an increase in the proportion of G1 phase cells, a decrease in cell size, a decrease in mitotic index is seen.
dsRNA made from templates generated with primers directed against this gene tested in RNAi screen for effects on Kc167 and S2R+ cell morphology.
Df(3L)ED4079 removes the first 2kb of one of the Pk61C transcripts and misses the other transcripts by 54bp.
Pk61C is an upstream effector of S6k.
Pk61C regulates cell growth and apoptosis via the Pi3K92E-dependant pathway.
