Bub3 functions in the "spindle assembly checkpoint (SAC)" pathway. This mitotic checkpoint pathway inhibits the APC/C complex and hence the transition from metaphase to anaphase, as long as there are free kinetochores that are not yet attached to the mitotic spindle in the cell. Bub3 is localized at the kinetochore during mitosis with maximal levels observed at free kinetochores. Together with other SAC proteins it forms mitotic checkpoint complexes that bind and inhibit the APC/C. [Date last reviewed: 2016-06-23]

Additional Descriptive Data
Bub3 protein shows a dynamic, cell cycle-dependent distribution pattern. During mitosis in cycling larval neuroblasts, interphase cells show a diffuse, nucleoplasmic staining. Bub3 protein becomes clearly localized to the kinetochore regions of the condensed chromosomes during prophase and prometaphase. As the chromosomes move to the metaphase plate, the intensity of Bub3 staining diminishes significantly. Weak kinetochore staining can be seen in early anaphase but disappears late in anaphase and telophase. During meiosis in spermatocytes Bub3 protein localizes to the kinetochores in prometaphase. It is also seen at the kinetochore region of spermatocytes during prophase/prometaphse of the second meiotic division. During metaphase, kinetochore staining becomes undetectable and no specific staining is observed in anaphase or telophase. In both mitosis and meiosis, Bub3 binds to the kinetochore during prophase/prometaphase. When individual chromosomes have reached a stable bipolar orientation at the metaphase plate, Bub3 protein begins to dissociate from the kinetochore.

      FlyBase curator comment: the insertion in the "f03381" Exelixis line (PBac{WH}CAP-D2f03381) was originally assigned to the Bub3 gene in FBrf0179797, resulting in the "Bub3f03381" (FBal0162743) allele. However, FBrf0184340 shows that the insertion is actually within CAP-D2.
      Other Comments
      RNAi screen using dsRNA made from templates generated with primers directed against this gene results in chromosome misalignment on the metaphase spindle when assayed in S2 cells. This phenotype can be observed when the screen is performed with or without Cdc27 dsRNA.
