Positive Regulators of Notch Signaling Pathway -
The Notch receptor signaling pathway is activated by the binding of the transmembrane receptor Notch (N) to transmembrane ligands, Dl or Ser, presented on adjacent cells. This results in the proteolytic cleavage of N, releasing the intracellular domain (NICD). NICD translocates into the nucleus, interacting with Su(H) and mam to form a transcription complex, which up-regulates transcription of Notch-responsive genes. hbs transcript is first expressed in a narrow band along the dorsal surface of stage 5 embryos, extending 2/3 of the length of the embryo. This band broadens laterally while decreasing in length until confined to the dorsal furrow. By stage 8, hbs is still expressed in the dorsal furrow, as well as ventrally in mesectodermal cells. At stage 10, hbs is expressed in the visceral mesoderm in lateral clusters of cells that resolve into patches by stage 11. By late stage 11, hbs is expressed in somatic and visceral mesoderm. By stage 12, hbs is present in fusion competent myoblasts, but not in muscle founder cells. Somatic mesoderm expression stops by stage 14; as mesoderm expression decreases, epidermal expression begins. Epidermal expression at stage 12 is observed at segment boundaries and in lateral patches; by stage 14, this expression is confined to muscle attachment sites. Stage 14 expression is also observed in pericardial cells. In the CNS,hbs is expressed in a set of ventral midline cells posterior to the developing posterior commissures at stage 12; by stage 14, expression is restricted to 2-3 cells below the posterior commissures, and absent in these cells by stage 15. hbs transcript is expressed in a subset of exit glial cells from stages 12-15. Third instar larval expression is observed in cells posterior to the morphogenetic furrow of the eye disc, and in cell clusters in the morphogenetic furrow. Expression in the wing disc is observed along both sides of the presumptive wing margin, and at presumptive wing veins L3 and L4. hbs is expressed in concentric circles in the leg discs. hbs is expressed in the larval optic lobes.

hbs protein expression is upregulated in third larval instar eye discs in the morphogenetic furrow and in clusters as they emerge from the morphogenetic furrow. There is also a low level of expression in all cells. By rows 2-4, hbs protein was enriched around ommatidial preclusters and in all developing photoreceptors (including R3/R4). Expression is maintained at later stages of eye development. The data suggest that hbs protein is enriched apically at plasma membranes near junctions and is also present in early and recycling endosomes.
hbs and sns are observed primarily in primary pigment cells in a pattern complementary to rst and kirre.
Expression of the hbs protein was detected in stage 15 embryonic Malpighian tubule stellate cells.

    Mutants exhibit a phenotype in which the A/P cone cell contact is improperly maintained through the end of pattern formation.
    Embryos mutant for hbs or overexpressing hbs show a partial myoblast fusion block.
    hbs appears to function as a dose dependant regulator of myoblast fusion.
    hbs appears to be a component of the molecular machinery that facilitates both myoblast and myotube guidance. hbs may also be involved in cell-cell contact events in the eye, bristle sensory oprgan and wing development.
