Gene Summary

PROTEIN O-GalNAcYLATION -
Protein O-linked GalNAcylation is a post-translational modification that occurs in the Golgi apparatus. The synthesis of O-linked GalNAc glycans (also known as mucin-type O-glycans) begins with the attachment of a N-acetyl-galactosamine (GalNAc) via an α-glycosidic bond to the hydroxyl group of serine or threonine residues in secreted and membrane proteins; this monosaccharide O-glycan is known as the Tn antigen. The O-linked GalNAc can be extended by the addition of a galactose, forming the core 1 T antigen. A glucuronic acid can be further attached to the galactose resulting in the formation of a linear glycan. In D. melanogaster, Tn and T antigens are the most prevalent type of O-glycans and are involved in a broad range of processes, including cell recognition, adhesion and signaling. Catalyzes the initial reaction in O-linked oligosaccharide biosynthesis, the transfer of an N-acetyl-D-galactosamine residue to a serine or threonine residue on the protein receptor (PubMed:30158631). It can both act as a peptide transferase that transfers GalNAc onto unmodified peptide substrates, and as a glycopeptide transferase that requires the prior addition of a GalNAc on a peptide before adding additional GalNAc moieties (PubMed:30158631).

Domain

There are two conserved domains in the glycosyltransferase region: the N-terminal domain (domain A, also called GT1 motif), which is involved in manganese coordination and substrate binding and the C-terminal domain (domain B, also called Gal/GalNAc-T motif), which is involved in catalytic reaction and UDP-Gal binding.

The alpha subunit loop in the ricin B-type lectin domain regulates substrate specificity and modulates the substrate access to the active site. In isoform A, the alpha subunit loop is composed of positively charged residues and acts on negatively charged substrates. In isoform B, the alpha subunit loop is composed of negatively charged residues and acts on positively charged substrates.

Alternative splicing of the eighth coding exon of Pgant9 has important functional consquences. Form "A" uses the upstream alternative exon and form "B" uses the downstream exon. qPCR in wandering third instar larvae shows that Pgant9A transcripts are the predominant forms in midgut, hindgut, tracha, Malpighian tubule, and CNS while Pgant9B transcripts are more abundant in salivary gland.
