Rbfox1 transcript is expressed in presumptive distal segments of the leg and antennal discs, and at the morphogenetic furrow of the eye disc.

Expression of Rbfox1 protein as detected by immunolocalization has the same pattern as the protein trap expression. It is first observed in the cytoplasm of early germline cysts. In germarium region 3, expression shifts from the cytoplasmic to the nucleus of germline cells where it persists until the end of oogenesis. Expression is observed in follicle cells beginning in germarium region 2b. Comparison to other germ cell markers shows that Rbfox1 has a novel expression pattern. It is observed in 4-cell, 8-cell, and 16-cell cysts and bridges the gap between early and late germ cell differentiation markers.
Rbfox1 protein is expressed in all imaginal discs. In the wing disc, expression is observed in the wing pouch and in myoblast cells (adepithelial cells) of the presumptive notum. Rbfox1 protein is expressed in the central regions of the antennal and leg discs, and in cells posterior to the morphogenetic furrow in the eye disc. Rbfox1 is also expressed in the CNS, PNS, and pericardial cells starting at the extended germ band stages of embryogenesis through larval stages, and in larval trachea and muscle cells. Rbfox1 protein is primarily localized to the nucleus in all tissues examined.
