Ank2 isoform Ank2-PL is expressed in the axon and presynaptic bouton at the neuromuscular junction. In the axon, expression of Ank2-PL is closely associated with the membrane and with the microtubule-associated protein futsch, and organised into a lattice-like structure. In the presynaptic boutons, Ank2-PL isoform is less abundant and found adjacent to the presynaptic active zone marker brp and in close proximity to the synaptic cell adhesion marker Fas2. In the areas between synaptic boutons, Ank2-PL protein is more abundant and organised into a lattice-like pattern.
The short isoform of Ank2 protein, i.e., Ank-PI, is localized to neuronal cell bodies, whereas antibodies to long isoforms stain primarily axonal pathways.
Different Ank2 isoforms show different subcellular localizations. They are expressed in the embryonic central and peripheral nervous systems. Ank2 is found at neuromuscular junctions both between and within synaptic boutons. It is expressed in periactive zones in neruomuscular junctions.
