BIR repeat containing ubiquitin-conjugating enzyme (Bruce) encodes a BIR domain containing anti-apoptotic protein of the inhibitor of apoptosis (IAP) protein family. It blocks apoptosis in part by binding to and ubiquitylating pro-apoptotic proteins such as the product of rpr. Bruce mutants are viable, but are vulnerable to apoptosis and show defects in spermatid differentiation. [Date last reviewed: 2019-03-21]

E2 UBIQUITIN CONJUGATING ENZYMES -
Ubiquitin-conjugating (E2) enzymes accept activated ubiquitin from ubiquitin-activating (E1) enzymes via thioester bond with a conserved catalytic cysteine. E2s then interact with ubiquitin ligases (E3) to directly or indirectly ubiquitinate target proteins. E2s are characterised by a 200 amino acid ubiquitin-conjugating (UBC) domain which interacts with E1 and E3s and acts catalytically. (Adapted from PMID:1994026).
INHIBITOR OF APOPTOSIS -
Inhibitor of APoptosis (IAP) proteins are defined by the presence of one or more BIR (baculoviral IAP repeat) domains. Not all IAPs inhibit apoptosis, but some interact with and inhibit caspases via their BIR domains. This interaction is displaced by RHG protein binding. (Adapted from FBrf0215539).

      dsRNA made from templates generated with primers directed against this gene tested in RNAi screen for effects on Kc167 and S2R+ cell morphology.
      Bruce can inhibit cell death induced by grim and rpr but not W.
      Alleles of this locus enhance the rpr and grim dependent eye phenotypes while most have little or no effect on the W induced phenotype.
