The following information accompanied stocks
donated to the Bloomington Stock Center by
Johannes Bischof and Konrad Basler, University of Zürich.
M{vas-int.Dm} carries a phiC31 integrase gene
that has been engineered to have the codon bias
of Drosophila melanogaster. Expression of the
integrase is driven by the vasa promoter. The
construction of M{vas-int.Dm} is similar to the
construction of M{vas-int.B} (FBtp0022219), which
was described in Bischof et al., 2007 (PNAS 104:
3312--3317;
http://flybase.net/reports/FBrf0199320.html).
Basically, the vasa promoter-phiC31 integrase
sequence, marked with Avic\GFP[E.3xP3]
(FBal0103092) and flanked by attB sites, was
integrated into M{3xP3-RFP.attP} via phiC31
integrase-mediated recombination. Other than the
altered integrase coding region, M{vas-int.Dm} is identical to M{vas-int.B}.
The presence both of Avic\GFP[E.3xP3] and
Disc\RFP[3xP3.PB] (FBal0193772) in M{vas-int.Dm}
causes RFP and EGFP expression in late pupal and
adult eyes. In practice, this leads to a pink eye
color from the RFP gradually appearing in adult
flies as they get older. This pink, however, is
clearly distinct from the color resulting from expression of white.
M{vas-int.Dm}ZH-2A is a homozygous and hemizygous
viable and fertile, X chromosome insertion at
2A. It was isolated by phiC31 integrase-mediated
recombination of the attB-vasa promoter-phiC31
integrase-attB sequence into M{3xP3-RFP.attP}ZH-2A.
M{vas-int.Dm}ZH-102D is a homozygous viable and
fertile, fourth chromosome insertion at 102D. It
was isolated by phiC31 integrase-mediated
recombination of the the attB-vasa
promoter-phiC31 integrase-attB sequence into M{3xP3-RFP.attP}ZH-102D.
In Bischof et al, 2007, the following insertions
from Supporting Information Table 4 are insertions of M{3xP3-RFP.attP}:

M{3xP3-RFP.attP}ZH-86Fb
M{3xP3-RFP.attP}ZH-102D

M{3xP3-RFP.attP} and M{3xP3-RFP.attP'} differ by only a few nucleotides.
