Subject: Microarray analysis of Df(2L)N22-14 Date: February 25, 2009 1:44:47 PM GMT-05:00 To: flybase-cambridgeXXXXXXXXXXXXXXX, yamamotoXXXXXXXXXXXXXXX, hbellenXXXXXXXXXXXXXXX, spanaXXXXXXXXXXXXXXX Microarray analysis of Df(2L)N22-14 Shinya Yamamoto, Eric Spana, Hugo Bellen and Kevin Cook DNA samples from Df(2L)N22-14 (FBab0022196) heterozygotes were compared to samples from wild type flies by Comparative Genomic Hybridization microarrays at the Duke Model System Genomics Unit as described in Erickson and Spana, 2006 (http://flybase.org/reports/FBrf0193934.html ). Corning CGAP slides spotted with the AROS Drosophila V1.1.1 ~70 nucleotide oligo set from Eurofins MWG Operon (www.operon.com <http://www.operon.com>) were used for the analysis. Most annotated genes were represented by a single oligo (denoted by a DM number). Sequences present at one copy in deletion heterozygotes are detected by lower relative fluorescence when compared to sequences present in two copies in wild type flies. _ _ The left Df(2L)N22-14 breakpoint lies in CG13386 or CG31898 or in the region between them, and lies in the range 2L:8399332..8401129 (R5) (predicted cytology: 29C1) based on the following evidence. _ _ The gene order at the left Df(2L)N22-14 end is CG13385 (FBgn0032039), CG13386 (FBgn0032040), CG31898 (FBgn0051898). A sequence in CG31898 (DM00015219, 2L:8401129..8401197 (R5)) was deleted. A sequence in CG13386 (DM00001838, 2L:8399264..8399332 (R5)) was not deleted, but CG13386 may have been disrupted. A sequence in CG13385 (DM00001837, 2L:8397274..8397342 (R5)) was not deleted. The right Df(2L)N22-14 breakpoint lies within CG13123 or nAcRalpha-30D or the region between them, and lies in the range 2L:9792596..9806483 (R5) (predicted cytology: 30D1) based on the following evidence. The gene order at the right Df(2L)N22-14 end is CG13123 (FBgn0032150), nAcRalpha-30D (FBgn0032151), FKBP59 (FBgn0029174), CG34183 (FBgn0085212), CG4537 (FBgn0032153). A sequence within CG13123 (DM00001618, 2L:9792528..9792596 (R5)) was deleted. A sequence within nAcRalpha-30D (DM00007315, 2L:9806483..9806531 (R5)) was not deleted, but nAcRalpha-30D may have been disrupted. A sequence within FKBP59 (DM00007599, 2L:9889462..9889530 (R5)) was not deleted.
