The reporter is indeed the 490 bp fragment from Nussbaumer et al. multimerized 4x and cloned into the minimal hsp70 promoter. The GAP is the transmembrane fragment from human GAP-43. I have the protein sequence (MLCCMRRTKQVEKNDEDQKI) for that, but not actually the DNA sequence. I need to make a new entry in FlyBase for this construct, and want to check I've got the details correct. I hope you can help. Looks like you used the 490/500bp fragment from Nussbaumer et al., 2000 Mech. Dev. paper (FBrf0129999) that they called the 'terminal cell enhancer (TCE)', which is 2.5kb upstream of the blistered ORF. This was multimerized, and then "used to drive expression of GAP::GFP ". Can you confirm the above, and clarify what is meant by "GAP". Thanks for your time, Steven Marygold FlyBase
