The following information accompanied stocks donated to the Bloomington Drosophila Stock Center by Bruce Baker, Howard Hughes Medical Institute, Janelia Farm Research Campus.
P{UAS-ix.IR} was made using a 484-bp fragment of the ix gene (coordinates  2R:11359517-11360000  in Dmel r6.13) amplified from Canton-S genomic DNA using these two primers (which add SpeI sites):
AAAAACTAGTCAGTGCAGCATCAACAGCAGC
AAAAACTAGTTATCAATCAGCCTGCGAAATGTT
The SpeI-digested product was inserted into the NheI site of pP{WIZ} (http://flybase.org/reports/FBmc0003001.html), and then again into the AvrII site of the resulting plasmid, so that the ix sequence is present as an inverted repeat. The orientation of the repeats is tail to tail (i.e., the order of the construct is UAS - hsp70TATA - <----ix---- - white intron - ----ix----> - SV40term). Tested by crossing to esg-GAL4 driver which caused sex transformation.
P{lexAop(FRT.Stop)mCD8.GFP} was made in a similar way to P{lexAop(FRT.Stp)tdTomato.nls}
