ttv[00681b] Ext2[1.8.1] double heterozygotes exhibit increased median survival time compared to controls when infected with group B Streptococcus (both wild-type and GAG-binding impaired mutants) and lower colony counts after infection.
ttv[00681b] Ext2[1.8.1] double heterozygotes exhibit reduced susceptibility to infection with the A909 strain of group B streptococcus.
Expression of ttv[Scer\UAS.T:Hsap\MYC] with Scer\GAL4[G14] leads to rescue of ttv[00681b] mutant larvae to adulthood or late pupal stages. The number of boutons per unit muscle area and bouton budding are increased to normal levels. Expression of ttv[Scer\UAS.T:Hsap\MYC] with Scer\GAL4[elav-C155] improves viability of ttv[00681b] mutant larvae. The number of boutons per unit muscle area are increased to normal levels while bouton budding is elevated above wild-type levels. Expression of ttv[Scer\UAS.T:Hsap\MYC] with Scer\GAL4[da.G32] fails to improve viability of ttv[00681b] mutant larvae but rescues the abnormally elevated mEJP frequency (but not the EJP amplitude) found in these larvae. The number of boutons relative to muscle area is significantly reduced compared with ttv[00681b] mutants. ttv[00681b] mutants exhibit morphological changes in somatic musculature, with mitochondrial distribution affected and altered Golgi organisation.
In response to nerve stimulation, ttv[00681b] mutants exhibit a significantly increased EJP compared with wild-type or heterozygous control larvae (20-25% increase). Spontaneous release events (mEJPs) are substantially reduced compared with heterozygous or wild-type controls, suggesting that loss of Heparan Sulfate biosynthesis affects the spontaneous vesicle release probability in the motoneuron. Post-synaptic mEJP amplitudes are significantly elevated in ttv[00681b] mutants relative to heterozygous or wild-type controls. ttv[00681b] mutant larval exhibit locomotion defects, with a significantly reduced number of peristaltic contractions per minute. ttv[00681b] mutant synapses are reduced in size and have significantly fewer boutons per unit muscle area compared with controls. ttv[00681b] motoneurons are bigger than normal, with a reduced number of \'buds\'. ttv[00681b] synapses consist primarily of large boutons without any buds. ttv[00681b] mutants exhibit an increase in activity-dependent endocytosis at the neuromuscular junction.
ttv00681b mutants display ISNb bypass at a similar rate to wild-type embryos.
Expression of hhScer\UAS.cIa with Scer\GAL4en-e16E in ttv00681b germ-line clone embryos doesn\'t rescue naked cuticle formation, but does increase denticle diversity in the ventral cuticle: denticles corresponding to row 2,3,4, and 5 types are all present.
ttv00681b germ-line clone embryos do not display any naked cuticle formation, although the resulting denticle lawn has some diversity.
ttvScer\UAS.T:Hsap\MYC with Scer\GAL4wg.PM completely rescues the cuticle phenotype of ttv00681b germ-line clone embryos.
ttv00681b/+; trol13/trol13 larvae have increased numbers of S phase larval neuroblasts per brain lobe than wild-type. Of ttv00681b/+; trolp4/trolp4 larvae, 29% show increased numbers of S phase neuroblasts per brain lobe and while 5% show decreased numbers.
Mosaic flies with eyes homozygous for ttv00681b (clones generated using the "eyFLP" system) have small eyes.
Homozygous mutant larvae have abnormally large boutons at the terminal arbor synapses of the ISN.
Homozygous larvae have a marked reduction in heparan sulphate levels, to at least 10-fold less than wild type. Heterozygotes show a slight reduction in heparan sulphate levels. Chondroitin sulphate is unaffected by ttv00681b.
ttv00681b/Df(2R)trix embryos derived from homozygous female germline clones show absence of naked cuticle.
Clonal analysis in the wing reveals that ttv is required in the receiving cell for the movement of hh gene product from the sending to the receiving cell.
Germline clones produce eggs with patterning defects: unrescued embryos exhibit mirror image duplication of denticle belts, rescued embryos are wild type. Lethality occurs during the larval and pupal stages.
