The addition of crol04418 enhances the wing blistering phenotype of if3/Y flies by 3-4 fold. The addition of crol04418 does not enhance the wing blistering phenotype of mys8/Y flies.
Homozygotes survive through embryogenesis and hatch to form first instar larvae. Homozygous larvae progress normally through prepupal development, showing no delay in puparium formation, but die before adult eclosion. Homozygotes die during two stages of pupal development. The first lethal phase occurs at the beginning of pupal development, during stage P5. Pupae that arrest at this stage have severe defects in head eversion and leg elongation. Approximately 40-50% of homozygotes die at stage P14, at the end of pupal development, with a microcephalic phenotype and malformed legs. The penetrance of the microcephalic phenotype is variable, while the "crooked legs" phenotype is highly penetrant and similar in all legs examined. The third pair of legs are smaller than wild-type, have a kink near the middle of the femur and lack some bristles in hemizygotes. Hemizygous leg discs show a range of defects from partial elongation to severe malformation. Legs showing partial elongation also have slightly more expanded 2nd to 5th tarsal segments compared to wild-type legs. Imaginal discs are normal in size and morphology.

Additional Information
Statement
Reference
Reporter gene expression is observed in leg imaginal discs isolated from late third instar larvae. Expression is initially restricted to the precursors of the tarsal segments. In prepupal leg discs, the expression expands to the precursors of the tibia and the femur. Expression is induced in the salivary glands at puparium formation. Expression is also observed in the fat body and in the trachea. Expression is observed in the ventral ganglion and in the presumptive optic lobes of the CNS in mid-third instar larvae and increases noticably at puparium formation. Finally, expression is observed in the corpus allatum of the ring gland.
In early embryo, expression in an 'anterior ring of cells'; expression in mandibular segment not certain.
In wandering third instar larva, no detectable β-gal staining.
