Clones of cells in the wing disc co-expressing Mmus\Ccnd1[Scer\UAS.cDa] and Hsap\CDK4[Scer\UAS.cDa] under the control of Scer\GAL4[Act5C.PP] show a 60-70% increase in median clone size compared to wild-type controls. Hph[02255]/+ suppresses this phenotype.
Hph02255/+ partially suppresses the increase in size of ommatidia and interommatidial bristles seen in seen in the eyes of Cdk4Scer\UAS.cMa; CycDScer\UAS.cDa; Scer\GAL4GMR.PF flies. FACS analysis of dissociated eye discs from Cdk4Scer\UAS.cMa; CycDScer\UAS.cDa; Scer\GAL4GMR.PF indicates that Scer\GAL4 expressing cells are larger than non-expressing and a decreased proportion are in S-phase. The increase in cell size, but not the reduction in S-phase, is partially suppressed by Hph02255/+. FACS analysis of dissociated eye discs from Cdk4Scer\UAS.cMa; CycDScer\UAS.cDa; Scer\GAL4GMR.PF indicates that Scer\GAL4 expressing cells are larger than non-expressing. This increase in cell size is partially suppressed by Hph02255/+.
Most Hph02255/Hph02255 or Hph02255/Df(3R)3-4 animals maily die during embryogenesis, but a few hatch into larva with severe growth defects that die die within 2 to 3 days. HphS030304/Hph02255 animals develop normally until pupariation, but very few escaper adults eclose. These escapers are smaller than their heterozygous siblings (18% lighter), but have normal body proportions. Cells in Hph02255 homozygous somatic clones (induced during embryogenesis) in the fat body of late third instar larvae are smaller and contain less DNA than heterozygous neighboring cells.
P{UAS-Hph.F}II.1; P{UAS-Hph.F}II.2; Scer\GAL4hs.PB rescues the lethality of Hph02255/HphS030304 even in the absence of heat shock. HphScer\UAS.cFa rescues the increase in ommatidial size in Cdk4Scer\UAS.cMa; CycDScer\UAS.cDa; Scer\GAL4GMR.PF suppressed by Hph02255/+.
