Expression of RanBPM[long.Scer\UAS] under the control of Scer\GAL4[elav.PLu] effectively rescues the response to light of RanBPM[k05201] mutant larvae. The overall locomotion of the mutant larvae is also improved. The feeding defects of RanBPM[k05201] mutant larvae are rescued by expression of RanBPM[long.Scer\UAS] under the control of Scer\GAL4[elav.PLu], Scer\GAL4[Mef2.PR] or Scer\GAL4[Mef2.247]. Expression of RanBPM[long.Scer\UAS] under the control of Scer\GAL4[Mef2.247] at 29[o]C significantly rescues the response to light of RanBPM[k05201] mutant larvae. The overall locomotion of the mutant larvae is also improved. Expression of RanBPM[S.Scer\UAS] under the control of Scer\GAL4[elav.PLu] effectively rescues the response to light of RanBPM[k05201] mutant larvae. The feeding defects of RanBPM[k05201] mutant larvae are rescued by expression of RanBPM[S.Scer\UAS] under the control of Scer\GAL4[elav.PLu], Scer\GAL4[Mef2.PR], Scer\GAL4[386Y] or Scer\GAL4[Mef2.247]. Expression of RanBPM[S.Scer\UAS] under the control of Scer\GAL4[Mef2.247] at 29[o]C significantly rescues the response to light of RanBPM[k05201] mutant larvae. Expression of RanBPM[S.Scer\UAS] under the control of Scer\GAL4[Mef2.PR] significantly rescues the response to light of RanBPM[k05201] mutant larvae.
Homozygous third instar larvae show a significantly lower response index (response index = (path length in dark - path length in light)/total path length during the assay) in an on/off light avoidance assay than control larvae. The mutant larvae show reduced locomotion and are uncoordinated (as seen by the equally high degree of direction change during the lights on and off pulses). RanBPM[k05201]/RanBPM[ts7] and RanBPM[k05201]/RanBPM[s135] third instar larvae show a significantly lower response index (response index = (path length in dark - path length in light)/total path length during the assay) in an on/off light avoidance assay than control larvae. Mutant larvae show reduced locomotion in constant darkness conditions. Homozygous larvae are smaller than control larvae of the same age from the early third instar stage onwards. A smaller fraction of homozygous early foraging third instar larvae that have been placed on a drop of yeast paste containing food dye after a 2 hour starvation period show food dye in the gut after 30 minutes on the yeast paste. If the feeding period after starvation is extended to 1.5 hours, the fraction of mutant larvae containing food dye in the gut at the end of the feeding period does not increase. In addition, a large fraction of the mutant larvae are found to have left the yeast drop (in contrast to wild-type larvae which mostly remain in the yeast drop). A smaller fraction of homozygous early foraging third instar larvae than control larvae move into and remain within a yeast drop if they are placed on medium outside a centrally located yeast drop containing food dye after a 2 hour starvation period and then allowed to feed for 1.5 hours (the control larvae largely move into and remain in the yeast drop in this assay). In addition, a lower proportion of the mutant larvae show food dye in the gut than control larvae at the end of the feeding period.
The increased number of branches and of boutons at the neuromuscular junction of Fmr1[EP3517] and Fmr1[Δ50M] larvae is significantly suppressed by RanBPM[k05201]/+.
