Construct: The complete 600bp zen ventral repression element (VRE) is fused upstream of a minimal 480bp eve stripe 2 enhancer (MSE), which drives expression of an eve-Ecol\lacZ fusion gene.
600bp zen ventral repression element (VRE) is fused upstream of eve MSE.
600bp XbaI-HindIII fragment carrying the complete zen ventral repression element (VRE) from position -1.6 to -1.0 is fused upstream of eve MSE.
Ecol\lacZ gene expression is driven by zen-eve promoter region fusion.

4.15kb SpeI fragment of the w gene that carries a deletion of a large part of the first intron and the regulatory region is reduced to 300bp preceding the transcription start site.

In stage 5 embryo, the ventral portion of the eve second stripe pattern is repressed; in addition, there is dorsal expression outside the eve second stripe. This results, during embryonic cycle 14A, in dorsal expression from the region of eve stripe 2 and more posterior; and during embryonic cycle 14B, in high levels of expression in the dorsal half of the eve stripe 2 region, with lower levels in more posterior dorsal regions.
