In EcollacZ-reporter constructs driven by either the -324 to +137 or the -117 to +137 Sry-β sequence (+1 is the Sry-β transcription start site), expression is detected at high levels in the germarium and nurse cells from stage S1 to S7 of oogenesis. Follicle cells begin to stain at stage S7, and by stage S10, follicle cell staining is strong, while expression is faint in nurse cells and oocytes. In adult males, EcollacZ staining is detected in the male accessory gland, the ejaculatory duct, and in the non-apical portions of the testi(suggesting that expression begins in late premeiosis). Staining is also seen in larvae, including larval tissue, the gonads of both sexes, and imaginal tissue.
