Description
The 48bp FRT sites that flank the w+mPB.FRT marker in the PBac{PB} element can efficiently recombine in the presence of FLP recombinase, removing the marker and regenerating a single 48bp FRT site. However, the 48bp site is significantly less efficient for longer distance recombination reactions than the longer FRT sites commonly used in constructs designed for generation of chromosomal deficiencies.
PBac{PB} comprises a complete piggyBac element with the open reading frame interrupted by a w+mPB.FRT marker flanked by short (48bp) FRT sites.

The w+mPB.FRT marker in the PBac{PB} element is flanked by 48bp Scer\FRT sites. Upon exposure to Scer\FLP1, these FRT sites efficiently recombine; the w+mPB.FRT marker is removed and a single 48bp FRT site is regenerated. The 48bp FRT site is significantly less efficient for longer distance recombination reactions than the longer FRTs commonly used in constructs designed for generation of chromosomal deficiencies.
