(Golic and Lindquist, 1989) Expression of a fragment of w genomic DNA with a 2.2kb deletion of 3' flanking sequences and a reduction in the size of the largest intron to 356bp is driven by a 388bp fragment of an Hsp70 gene. 	The let-7 seed sequence (GAGGTAGTAGGTT) has been deleted from the progenitor construct. (Sokol et al., 2008) The P{W8,let-7-C[Δlet-7]} construct can rescue the male fertility and climbing defects of Df(2L)let-7-C[KO1]/Df(2L)let-7-C[GKI] adults. However, the rescued adults still show severely reduced locomotion and show partial defects in flight. Df(2L)let-7-C[KO1]/Df(2L)let-7-C[GKI] females rescued by P{W8,let-7-C[Δlet-7]} show severe defects in female fertility and oviposition. (Sokol et al., 2008) Df(2L)let-7-C[KO1]/Df(2L)let-7-C[GKI] adults rescued by the P{W8,let-7-C[Δlet-7]} construct still show abnormal persistence of the dorsal internal oblique muscles (these muscles decay during post-eclosion maturation in wild-type animals) and still show defects in the dorsal muscles and their neuromuscular junctions. (Sokol et al., 2008)
