The snRNA:U7 gene, including the putative 3' processing signals, are cloned upstream of GFP coding sequences, followed by a strong canonical polyA sequence, downstream of the stop codon. Constitutes a reporter for snRNA misprocessing; under normal conditions, the construct should produce a properly processed snRNA:U7 snRNA and GFP should not be expressed. However, if snRNA processing is impaired, since the snRNA:U7 sequences prior to the GFP ORF do not contain a AUG start codon, the resultant mRNA is expected to become polyadenylated, encode a functional GFP protein, and contain the snRNA:U7 sequence as its 5'UTR.
