l(3)mbtGM76/Df(3R)ED10966 females do not lay eggs; ovaries are atrophied and ovarioles contain fewer individualized egg chambers than l(3)mbtGM76 heterozygous controls; egg chambers occasionally contain apoptotic cells and are highly abnormal, showing follicle layer integrity defects, accumulation of undifferentiated germ cells (i.e. marked by branched fusomes and enclosed by follicle cells), extra nurse cells and oocytes, egg chamber fusion (e.g. occasionally there are multiple germaria connected to the same aberrant egg chamber) and accumulation of stalk cells on top of follicle cells, as compared to l(3)mbtGM76 heterozygous controls; however, similar to controls, mutant germaria present germline stem cells adjacent to the somatic niche.

Similar to controls, l(3)mbtGM76/Df(3R)ED10966 transheterozygous ovaries in L3 have distinct apical compartments harboring terminal filaments, as well as a medial region containing germ cells and intermingled cells; however, there is a decreased number of somatic cells in a similarly sized germ cell containing region and mutant intermingled cells fail to migrate in between the progenitor germline cells and instead surround them.

Egg chambers bearing l(3)mbtGM76 homozygous follicle cell clones show supernumerary nurse cells or multiple oocytes.

l(3)mbtGM76/Df(3R)ED10966 females that also express l(3)mbtUASp.Tag:MYC under the control of Scer\GAL4tj-NP1624 produce embryos that fail to hatch.

The volume of the optic lobe neuroepithelium is significantly increased in size in homozygous and l(3)mbtGM76/Df(3R)D605 third instar larvae compared to wild type. Cell number and mitotic pattern within the optic lobe appears normal in homozygous first and early second instar larvae, but starting in late second instar, the mutant epithelia expand and start folding.

Embryos resulting from l(3)mbtGM76 germ-line clones have reduced numbers of germ cells compared to wild-type embryos.

The ovariole/egg chamber defects observed in l(3)mbtGM76/Df(3R)ED10966 females are weakly and strongly suppressed by pum680 heterozygosity and homozygosity, respectively.

At 25[o]C temperature, Lint-11/Lint-11, l(3)mbtGM76/+ females are sterile, unlike Lint-11/Lint-11 or l(3)mbtGM76/+ females.

The increase in volume of the optic lobe neuroepithelium which is seen in l(3)mbtGM76 homozygotes is suppressed if the larvae are also homozygous for Df(3L)banΔ1.

In trans Df(3R)D605 or Df(3R)mbtPE3, l(3)mbt alleles form the following phenotypic series: l(3)mbtGM161 < l(3)mbtts1 < l(3)mbtGM79 < l(3)mbtE2 < l(3)mbtGM76. Phenotypes range from weak maternal-effect mitotic asynchrony to a severe mitotic block to oogenesis defects in trans-heterozygous females. Each of these phenotypes is temperature sensitive: phenotypes increase in severity as the temperature is raised from 18 to 29oC.
